Course Description:
The course is a specialized core course for graduates in department of chemistry, biology and other related areas. Modifying and labeling molecules at will, is always chemists’ goal. Molecular modification and chemical labeling, as significant experimental techniques, are playing an important role in chemistry, medicinal chemistry, biomedical, biochemistry, biotechnology and molecular biology. They are indispensable for graduates and research specialist staffs. The course gives a comprehensive and deep-going introduction of the basic theory, general situation, technology and development of molecular modification and chemical labeling. The course firstly presents basic knowledge of biochemistry, chemical synthesis, fluorescence and radiochemistry, then the basic approach and technology of molecular modification and chemical labeling, with living examples to demonstrate the application of these techniques in scientific research practice. Students are expected to gain an intimate knowledge of the rapid development of molecular modification and chemical labeling and grasp the basic theory principle and methods of molecular modification and chemical labeling, which have been used more and more extensively, thus ram a specialized rationale for their scientific research. The course is suitable for graduates and research specialist staffs as an optional course.
Course Content:
Chapter 1 Introduction Intercross fusion and interinfiltration between chemistry and biology; Origin, basic concepts and advances of molecular modification and chemical labeling, etc. Chapter 2 Biochemical basis of molecular modification and chemical labeling Brief introduction of basic theory of biology: amino acid, peptide, protein, nucleotide, nucleic acid, peptide nucleic acid, DNA/RNA/genes, central dogma, biochemistry, biophysical process, etc. Chapter 3 Chemical synthetical basis of molecular modification and chemical labeling Introduction of basic methods and principles of chemical synthesis (inorganic synthesis, organic synthesis, biochemical enzymatic synthesis, etc); Introduction of chemical synthesis of peptide and oligonucleotides. Chapter 4 DNA molecular modification technology Introduction of frequently used and recently advanced methods and techniques of molecular modification: DNA terminal modification, DNA sequence modification, oligonucleotide-peptide combination, etc; Separation and analysis of modified products. Chapter 5 DNA molecular marking technology——nonradioactive marking Introduction of frequently used and recently advanced methods and techniques of nonradioactive marking, such as Biotin marker and EDTA marker; Separation and analysis of nonradioactive labeled products and probes. Chapter 6 Protein modification and labeling Introduction of frequently used and recently advanced methods and techniques of molecular modification: DNA terminal modification, DNA sequence modification, oligonucleotide-peptide combination, etc; Separation and analysis of modified products. Chapter 7 Molecular marking technology——radioactive marking Basic concepts of radiochemistry and radiation protection; Basic methods and techniques of radiochemical labeling; Radioactive metallic element labeling and radioactive nonmetallic element labeling; Separation and analysis of radiolabeled products and probes. Chapter 8 Application of molecular modification and molecular labeling Brief introduction of medicinal chemistry; Different sorts of pharmaceutical compounds; The process and method of new drugs research; The application of molecular modification in biotechnology, medicine development, pharmacological research, clinical diagnosis and uses in treatment.
